The project starts with SMART™ cDNA production. The stored full-length SMART™ cDNA library is the key source for template preparation and downstream applications such as individual cDNA isolation, library plating and sRNA transcription.

Using proprietary procedures, the SMART™ cDNA is converted into new adapter-tagged cDNA at the 5’-ends and the templates are purified from unligated adapters and fragmented into small 400-600bp fragments. The templates are enriched for 5’-ends and 3’-end adapters are ligated according to 454 protocols.

454 Life Sciences will perform emPCR and GS FLX Sequencing using Titanium chemistry. The result will be 200-600k fragments of maximal 400nt in length.



As of mid-2008, GreenAcres works with CLC Bio software for analysis and annotation. In addition to the 454 data, the recently launched CLC Genomics Workbench is capable of integrating the de novo assemblies of Sanger, Solexa, Helicos, and SOLiD reads and supports a large number of downstream data. GreenAcres and CLC Bio are developing proprietary software that manages reads of 5’-ends with known adapter sequences on either end.
